Prepared microscope slide: Hamster adrenal gland Set of 3
All views of these slides are at 40x. These are all fixed in Bouin’s fluid They are then stained with either Harris’ Hematoxylin or Heidenhain’s Hematoxylin. Then they are counter-stained. Samples are 10μ thick. The main slide shown is stained with Harris’ Hematoxylin and counter-stained with Methyl green. There are 2 additional slides in the set. The first is stained with Harris’ Hematoxylin counter-stained with Orange G. The second is stained with Heidenhain’s Hematoxylin and counter-stained with Eosin. They may be seen by clicking either of the following lines.

These slides were prepared sometime prior to the 1950s by an individual named Kirschbaum.
Bouin’s fluid contains picric acid, which has the property that it renders tissue more readily stained by acid dyes. Harris’ Hematoxylin is a nuclear stain that produces a deep purple blue. Heidenhain’s Hematoxylin uses two solutions, the first being an iron mordant such as ferric chloride or ferrous ammonium sulfate followed by an application of hematoxylin.
